DNA tailing without terminal transferase.

چکیده
Tailing of DNA with terminal transferase is a technically demanding, but essential step in full-length cDNA cloning procedures*•^. It is also an essential step of a poly(dA) affinity labelling technique, which has been used to select families of partial restriction fragments on oligo(dT)cellulose^•*. In order to bypass the need for transferase in the latter technique, we made a simple modification of DNA-linker design, to produce a "tailing-adapter" which may have broader applications. The top strand of the tailing-adapter (A), was synthesised and 5' phosphorylated with T,j-kinase. It was then

Controlled ribonucleotide tailing of cDNA ends (CRTC) by terminal deoxynucleotidyl transferase: a new approach in PCR-mediated analysis of mRNA sequences.
Controlled ribonucleotide tailing of cDNA ends (CRTC) by terminal deoxynucleotidyl transferase is a polymerase chain reaction (PCR)-mediated technique that was developed to facilitate cloning and direct sequence analysis of complete 5'-terminal unknown coding regions of rare RNA molecules. In contrast with standard tailing protocols using dNTPs as the substrate, ribo-tailing of cDNA ends is eas...
متن کاملCharacterization of the extreme 5' ends of RNA molecules by RNA ligation-PCR.
1Permanent address: P..O. Box 677, Nedlands, Western Australia, 6009. Despite major recent advances in PCRbased methodology, characterization of the extreme terminal sequences of RNA molecules may be difficult and time consuming, particularly when the 3' end is not polyadenylated.~l~ The usual approach to defining the 5' end involves cDNA synthesis using gene-specific downstream priming of the ...
متن کاملTerminal transferase-dependent PCR: a versatile and sensitive method for in vivo footprinting and detection of DNA adducts.
We report here a new, sensitive and versatile genomic sequencing method, which can be used for in vivo footprinting and studies of DNA adducts. Starting with mammalian genomic DNA, single-stranded products are made by repeated primer extension; these products are subjected to homopolymeric ribonucleotide tailing at the 3' termini with terminal deoxynucleotidyl transferase and then ligated to a ...
متن کاملTailing cDNAs with terminal deoxynucleotidyl transferase in RT-PCR assays to identify ribozyme cleavage products
Polytailing a cDNA with terminal deoxynucleotidyltransferase (TdT) results in the addition of a homopolymeric sequence at its 3'-end. Here we describe the use of tailing in competitive RT-PCR assays to evaluate cleavage efficiency of ribozymes. Using a system that perfectly mimics intracellular cleavage, we were able to detect as few as 1% of cleaved moieties. Furthermore, employing primers ove...
متن کاملTrack detection on the cells exposed to high LET heavy-ions by CR-39 plastic and terminal deoxynucleotidyl transferase (TdT)
Background: The fatal effect of ionizing radiation on cells depends on Linear Energy Transfer (LET) level. The distribution of ionizing radiation is sparse and homogeneous for low LET radiations such as X or γ, but it is dense and concentrated for high LET radiation such as heavy-ions radiation. Material and Methods: Chinese hamster ovary cells (CHO-K1) were exposed to 4 Gy Fe-ion 2000 keV/...
